Microspheres of chitosan-bioactive glass for application in orthopedic surgery. In vitro experiment.

Résumé

In order to increase the biocompatibility and bioactivity of bioactive glass, a biodegradable therapeutic polymer (Chitosan: CH) has been incorporated into bioactive glass (BG) by freeze-drying to synthesize biocomposite microsphere material for applications in bone surgery. The interfacial links between bioactive glass and chitosan polymer were investigated though Fourier transformed IR absorption spectroscopy (FTIR). The surface morphol. of the biocomposite and the homogeneous dispersion of bioglass particles in the matrix of chitosan polymer were examd. by SEM. The bioactivities of the BG and of the BG/CH biocomposite microsphere were investigated by soaking the powder samples in a simulated body fluid (SBF). The formation of a biol. active hydroxycarbonate apatite (HCA) layer on the surfaces of BG and also BG/CH biocomposite was detected. ICP-OES method was employed to evaluate the rate of bioactivity of these two biomaterials. The obtained results indicate that the chitosan enhances the bioactivity of bioactive glass. The biocomposite based on bioactive glass and chitosan polymer have an excellent ability to form an apatite layer on its surface. After three days of immersion, FTIR analyses confirm the formation of this layer. The cytotoxicity and cell viability assays were also investigated by the MTT method using osteoblast like cells SaOS2. The results confirm the absence of toxicity and a good cell proliferation on this biomaterial. However, the addn. of chitosan in the glass matrix enhances the cell proliferation of about 5% at 6 days after culturing.
